Genetic Manipulation of H. pylori

Genetic manipulation is an important tool to investigate various genes and proteins of H. pylori. Genetic manipulation of H. pylori through genetic engineering and selective breeding can lead to different strains of H. pylori, which can be used in studies to discover the pathogenicity, virulence, and vulnerability to survival, thus providing solutions for the prevention and treatment of H. pylori infections. To facilitate the study of H. pylori, Ace Therapeutics offers two genetic manipulation services for H. pylori, including the selective culture of H. pylori strains and deletion or overexpression of specific H. pylori genes.

Selective culture of H. pylori strains

Ace Therapeutics has dedicated H. pylori culture staff to isolate and culture H. pylori. They have long training in culturing and can isolate and culture H. pylori from stomach biopsies, saliva, dental plaque, and stool.

Although the cultural conditions for H. pylori are demanding, we take several measures to ensure the efficient culture of H. pylori. Firstly, H. pylori need to grow on nutrient-rich media, so we add high-quality sheep blood, horse blood, and our specially formulated H. pylori nutritional additive to the H. pylori culture medium. Then, H. pylori need to be grown in a micro-oxygenated environment with a high concentration of CO2 (5% to 10%) and high humidity (98%) for a long period of time (5-7 days). Therefore, we use special H. pylori incubators to better control the cultural environment. What's more, most H. pylori strains have spontaneous mutation rates between 10-5 and 10-7, which is several orders of magnitude higher than the average mutation rate of Escherichia coli. Therefore, we pay close attention to mutations during the culture of H. pylori and we have good methods to monitor the variation.

Gene deletion or overexpression of specific H. pylori genes

Gene deletion or overexpression of specific H. pylori genes facilitates characterization and functional analysis of individuals. Ace Therapeutics offers gene deletion or overexpression services of specific H. pylori genes to support H. pylori studies. Not only do we provide H. pylori single-gene deletion or overexpression services, but we also provide multiple gene deletions or overexpressions to facilitate in-depth studies by investigators.

Take the following as an example. In studying the pathological and physiological role of HofF, the outer membrane protein encoded by the H. pylori gene hp0788, we offer deletion or overexpression services of the hp0788 gene to obtain a new H. pylori strain (variant hp0788). Experimenters can then perform various controlled experiments using the original H. pylori strain and the variant hp0788 to explore the physiological role of HofF and its impact on pathogenesis. In addition, depending on the results of those studies, we provide further knockdown or overexpression services of the relevant genes to explore the relationship between other genes and hp0788 in more depth.
